Rheumatoid arthritis (RA) is a chronic autoimmune disease where methotrexate (MTX) is the standard first-line therapy, though patient response and tolerability can vary with dose. To address this variability, a post hoc analysis of the ORAL Strategy study evaluated the efficacy and safety of tofacitinib and adalimumab across different baseline and weight-adjusted MTX doses in 1146 patients with active RA. The study found that ACR50 response rates at 6 months were similar across BMI-normalized MTX dose tertiles in both tofacitinib and adalimumab combination therapy groups, with comparable trends seen in weight-normalized analyses.
However, responses were numerically higher with combination therapy than with tofacitinib monotherapy. No consistent relationship was observed between MTX dose and safety outcomes. These findings suggest that tofacitinib demonstrates consistent efficacy and noโฆ
