Several HER-kinase inhibitors and HER2-antibody treatments have demonstrated the successful inhibition of HER2 in HER2-amplified breast cancer. However, resistance to anti-HER2 drugs poses a challenge in treating metastatic breast cancer. Nevertheless, the discovery and pharmacologic inhibition of the HER2 oncogene in breast cancer has had a significant impact on targeting and treating breast cancer. Several anti-HER2 drugs have been developed and tested in the past few years, with three new drugs approved in 2020 alone.
