Overexpression of human epidermal growth factor receptor 2 (HER2) triggers several pathways that result in the proliferation of breast cancer cells. About 15–20% of breast cancers overexpress HER2, which makes it an optimal target for HER2-amplified breast cancer treatment. However, resistance to anti-HER2 drugs remains a challenge in metastatic breast cancer. Nevertheless, the discovery and pharmacologic inhibition of the HER2 oncogene in breast cancer represents a hallmark success in targeted therapy in oncology. Several anti-HER2 drugs have been developed and tested in the past few years, with three new drugs approved in 2020 alone.
