Clozapine was synthesized in 1956 and the structure was based on the chemical structure of the tricyclic antidepressant imipramine. This drug was discovered only six years after the first-generation drug, chlorpromazine. Recently, the FDA has strengthened its warning against clozapine for causing constipation, leading to serious bowel obstruction. Introduction : The U.S. Food and Drug Administration has recently raised a warning regarding serious complications caused by clozapine viz constipation.
The FDA has further strengthened this warning stating that untreated constipation can cause serious issues and obstruction in the bowel.
