The Phase 3 RHONE-X extension (NCT04432831) evaluated long-term safety and exploratory efficacy of faricimab, a dual Ang-2/VEGF-A inhibitor, in patients with DME who completed YOSEMITE/RHINE. Among 1,474 participants transitioned to a treat-and-extend (T&E) regimen without reloading doses, 82% completed the 2-year extension. Faricimab maintained visual gains ( ≈+9.5 to +11.4 ETDRS letters ) and robust CST reductions (≈ –198 to –205 μm ) from original baseline across prior treatment groups, including aflibercept. Over 90% achieved absence of DME (CST <325 μm) at study end.

Treatment burden remained low, with a median of 7–8 injections over 2 years and ~80% maintained at ≥Q12W intervals.
