Prurigo nodularis (PN) is a chronic inflammatory skin disease characterized by intensely pruritic papulonodular lesions, and many patients have coexisting atopic conditions. Dupilumab is the first approved therapy for PN; however, its efficacy in patients with and without atopic comorbidities remains poorly established. To address this gap, a study evaluated the efficacy and safety of dupilumab in 311 patients with PN with or without a history of atopic comorbidities.

The study found that significantly more patients treated with dupilumab achieved clinically meaningful improvements compared with placebo at week 24 in both atopic and non-atopic subgroups for itch (58.2% vs 20.6% and 59.3% vs 17.8%), clear or almost clear skin (52.2% vs 16.2% and 41.9% vs 17.8%), and combined itch and skin lesion outcomes (37.3% vs 7.4% and 33.7% vs 10.0%).
