Prurigo nodularis (PN) and chronic pruritus of unknown origin (CPUO) are chronic pruritic disorders that substantially impair quality of life, yet therapeutic options remain limited. To address this gap, a study evaluated the efficacy and safety of 200-mg oral abrocitinib, a Janus kinase 1 inhibitor, administered once daily in adults with moderate to severe PN or CPUO, involving 25 participants. The study found that treatment with 200-mg oral abrocitinib once daily led to substantial reductions in pruritus, with mean (SD) baseline peak pruritus numerical rating scale (PP-NRS) scores of 9.2 for PN and 8.2 for CPUO.
