Daratumumab has been approved for multiple myeloma treatment. However, its safety and efficacy in treating high-risk smoldering multiple myeloma, a precursor disease of active multiple myeloma, remain unclear. Now, a phase 3 clinical trial has investigated the effectiveness of daratumumab in treating 390 patients with high-risk smoldering multiple myeloma . Over a 65.2-month follow-up period, the study found that daratumumab lowered the risk of disease progression or death by 51% compared to active monitoring.
At five years, 63.1% of patients in the daratumumab group remained progression-free, compared to 40.8% in the active monitoring group. Similarly, overall survival at five years was significantly higher in the daratumumab group than in the active monitoring group (93.0% vs. 86.9%).
