Upadacitinib, an oral selective Janus kinase inhibitor approved for moderate-to-severe atopic dermatitis (AD), requires further long-term evaluation beyond one year. To address this need, a large international clinical study assessed the extended efficacy and safety of upadacitinib over 140 weeks in patients with moderate-to-severe AD, involving 2,782 participants. The study showed that long-term treatment with upadacitinib produced durable clinical benefits, with high proportions of patients maintaining Eczema Area and Severity Index (EASI) 75/90 and validated Investigator Global Assessment for AD score of clear/almost clear (vIGA)-AD 0/1 responses through week 140.
