A phase 3 clinical trial evaluated the safety and efficacy of the novel oral GLP-1 receptor agonist orforglipron compared with oral semaglutide in 1,698 adults with type 2 diabetes inadequately controlled on metformin. The 52-week study was conducted across 131 research centres and hospitals in Argentina, China, Japan, Mexico, and the USA. At week 52, HbA1c levels decreased from a baseline of 8.3% by 1.71% in patients receiving orforglipron 12 mg and by 1.9% in those receiving orforglipron 36 mg, compared with reductions of 1.23% and 1.47% in the semaglutide 7 mg and 14 mg groups, respectively.
