Mantle cell lymphoma (MCL) is a rare but severe form of non-Hodgkin lymphoma . The long-term prognosis is generally poor and requires aggressive treatment strategies to manage this disease better. The USFDA recently approved the first non-covalent Bruton’s tyrosine kinase (BTK) inhibitor named pirtobrutinib for patients with relapsed or refractory MCL in adults, particularly those who had been treated with covalent BTK inhibitors.

The approval was based on the results of a phase 1/2 open-label, multicentric single-arm study, which consisted of 120 MCL patients who had previously undergone treatment with two lines of systemic therapy, including at least once with a BTK inhibitor.
